Fast and cAMP-Sensitive Mode of Ca -Dependent Exocytosis in Pancreatic -Cells

چکیده
The fast component (mode 1) of Ca -dependent exocytosis in pancreatic -cells, unlike that in adrenal chromaffin cells, is regulated by cytosolic ATP in a concentration-dependent manner. This action of ATP is apparent within 3 min and does not require ATP hydrolysis; rather, it requires the production of cAMP by adenylate cyclase. Moreover, the effect of cAMP is ATP dependent, as revealed by the observation that the fast component of exocytosis is facilitated by ATP, even in the presence of a saturating concentration of cAMP (200 mol/l). Thus, the amplitude of mode-1 exocytosis depends quadratically on the cytosolic ATP concentration and is facilitated by ATP, even in the absence of an increase in the concentration of cAMP. Given that high glucose concentrations increase the cytosolic ATP concentration, glucose-induced insulin secretion likely involves this action of ATP on mode-1 exocytosis, together with its effect on ATP-dependent K channels. In contrast to the fast component of exocytosis, the slow component (mode 2) of this process is independent of cAMP and ATP and can account for the slow component of insulin secretion, which does not require these nucleotides. Cyclic AMP potentiates Ca2+-dependent exocytosis in pancreatic duct epithelial cells
Exocytosis is evoked by intracellular signals, including Ca(2+) and protein kinases. We determined how such signals interact to promote exocytosis in exocrine pancreatic duct epithelial cells (PDECs). Exocytosis, detected using carbon-fiber microamperometry, was stimulated by [Ca(2+)](i) increases induced either through Ca(2+) influx using ionomycin or by activation of P2Y2 or protease-activate...
متن کاملExtracellular calcium influx activates adenylate cyclase 1 and potentiates insulin secretion in MIN6 cells.
Intracellular cAMP and Ca(2+) are important second messengers that regulate insulin secretion in pancreatic β-cells; however, the molecular mechanism underlying their mutual interaction for exocytosis is not fully understood. In the present study, we investigated the interplay between intracellular cAMP and Ca(2+) concentrations ([cAMP](i) and [Ca(2+)](i) respectively) in the pancreatic β-cell ...
متن کاملPerspectives in Diabetes Epac: A New cAMP-Binding Protein in Support of Glucagon-Like Peptide-1 Receptor–Mediated Signal Transduction in the Pancreatic -Cell
Recently published studies of islet cell function reveal unexpected features of glucagon-like peptide-1 (GLP-1) receptor–mediated signal transduction in the pancreatic -cell. Although GLP-1 is established to be a cAMPelevating agent, these studies demonstrate that protein kinase A (PKA) is not the only cAMP-binding protein by which GLP-1 acts. Instead, an alternative cAMP signaling mechanism ha...
متن کاملCalcium dependencies of regulated exocytosis in different endocrine cells.
Exocytotic machinery in neuronal and endocrine tissues is sensitive to changes in intracellular Ca(2+) concentration. Endocrine cell models, that are most frequently used to study the mechanisms of regulated exocytosis, are pancreatic beta cells, adrenal chromaffin cells and pituitary cells. To reliably study the Ca(2+) sensitivity in endocrine cells, accurate and fast determination of Ca(2+) d...
متن کاملCyclic AMP dynamics in the pancreatic β-cell
Insulin secretion from pancreatic β-cells is tightly regulated by glucose and other nutrients, hormones, and neural factors. The exocytosis of insulin granules is triggered by an elevation of the cytoplasmic Ca(2+) concentration ([Ca(2+)](i)) and is further amplified by cyclic AMP (cAMP). Cyclic AMP is formed primarily in response to glucoincretin hormones and other G(s)-coupled receptor agonis...
